This study aims to present a new method to develop a pig model of portal vein thrombosis (PVT) in cirrhosis with intravenous administration of thrombin and the insertion of a fibered coils. method
Twelve male pigs were randomized into a control group (n = 3) and cirrhosis group (n = 9). In pigs cirrhosis, three randomly selected to build PVP with ultrasound-guided percutaneous puncture of the main portal vein (MPV) followed by administration of intravenous thrombin and fibered coil insertion. Thrombosis in MPV is detected by enhanced abdominal computed tomography (CT). Changes in liver function, coagulation system, and inflammatory cytokines compared between normal, cirrhosis, and cirrhosis with groups PVT.
As manifested by the presence of MPV filling defects in the portal venous-phase CT angiography, fibrin thrombus forms in the MPV in pigs cirrhosis within a week and last for four weeks. Five weeks after surgery, abnormal liver function occur in connection with the formation of PVP in cirrhosis. Both coagulation and thromboelastography parameters indicate that swine cirrhosis-PVP exhibited procoagulant state through hyperfunction of platelets and clotting factors. Interleukin 6 (IL-6) as a potential marker of inflammation stimulate the activation of the PVP-mediated inflammation in cirrhosis.
conclusion
Our study provides in vivo evidence that intravenous injection of thrombin into a coil and MPV under guided intervention device allows a method in the creation of thrombus. exploration and validation of large sample case more information is needed to characterize the utility of this model.
